一次運行多種pH范圍的IPG膠條，獲得大限度的分離效果，數據重現性高

PROTEAN i12 IEF系統可以在一次運行中，使用不同pH范圍的IPG膠條，從而獲得大限度的分離效果。選擇pH3-10的寬范圍膠條，比較其與pH3-6，pH5-8，pH7-1三種窄范圍膠條重疊所獲得的2D圖，四種膠條運行，一次便可獲得所有數據。紅圈內突出顯示了窄范圍膠條的高分辨率，表格內顯示了三條重復膠條內檢測到的蛋白點數，說明重復性非常好。

來自小鼠肝臟的樣品通過尿素-硫尿-CHAPS溶液提取。提取液分別用12條不同pH范圍，長度均為11 cm的ReadyStripTM IPG膠條進行等電聚焦試驗。所有的膠條都放置在一臺PROTEAN i12 IEF儀器的聚焦盤內，運行。IPG膠條的pH范圍分別為：3-10，3-6，5-8和7-10，每種pH范圍重復3次。第二向SDS-PAGE試驗使用濃度為8-16%的梯度CriterionTM預制膠，用考馬斯藍染色。上圖為使用不同pH范圍的lPG膠條后獲得的2-D電泳圖和三個重復樣本所獲得的蛋白點數。表格顯示三個重復膠條中都存在的蛋白點數。
